Abstract
Summary The long pentraxins PTX3 is involved in different pato-physiological contexts, ranging from innate immunity to female fertility. It binds to diverse ligands, such as the outer membrane protein A from Klebsiella pneumoniae (KpOmpA), the extracellular matrix component (TSG-6) and the angiogenic factor FGF-2. In innate immunity, PTX3 plays a non redundant role in resistance against selected pathogens. We explored in vivo the role of PTX3 in response to KpOmpA. According to our results, KpOmpA induces an inflammatory response, amplified by the co-administration of PTX3, in terms of leukocyte recruitment and cytokine production, through complement activation. In female fertility, PTX3 is a structural component of the extracellular matrix of the cumulus oophorus, together with TSG-6. Our data revealed that PTX3 and TSG-6 are co-regulated in leukocytes in response to pro-inflammatory mediators and co-localize in inflamed tissues.
