Abstract
Genetic adaptation of Pseudomonas aeruginosa to CF lungs establishes hypermutable and antibiotic resistance variants. Sara Montanari - Matricola R06355 P. aeruginosa lung infection is the major cause of morbidity and mortality in CF patients. During long-term colonization of the CF niche P. aeruginosa goes through a genetic diversification becoming mucoid, resistant to antibiotics and hypermutable. The outstanding ability of P. aeruginosa to adapt is supported by several mechanisms including gene regulation and adaptive mutation. The impact of anaerobic growth condition to P. aeruginosa genotypic and phenotypic changes was assessed by genome expression profile analysis. The 8% of P. aeruginosa genome (440 genes) resulted differentially expressed in response to anaerobiosis vs aerobiosis. 16 genes involved in antibiotic resistance were found significantly changed including oprD a major porin specific for imipenem influx. Down-regulation of oprD was confirmed by RT-PCR and reduced killing of bacterial colture was proven under anaerobiosis in vitro [...]
