Prostate cancer is among the most common cancer in men. To set new diagnostic and therapeutic approaches it is critical to have preclinical models resembling the human neoplasia and its progression mechanism. In order to understand the functions of two components of the extracellular matrix, osteonectin (SPARC) and osteopontin (OPN), in the progression and onset of prostatic desease, I have breeded the TRAMP (transgenic adenocarcinoma mouse prostate) model with OPN and/or SPARC knock out (KO)mice. Because of the difficulty to track the progression of this kind of tumor, these models have been studied with the ultrasound Visualsonic's Vevo 770 imaging system. Macroscopic observations evidentiated a more aggressive phenotype in comparison to parental strain, and immunohistochemical (IHC) analysis indicated that the tumor that arises in these KO models is a small cell carcinoma with neuroendocrine phenotype.
