Tyrosine nitration of cytoskeletal proteins in physiological conditions: role of nitrated alpha-tubulin in microtubule stability. NO is a signalling molecule implicated in a spectrum of cellular processes including neuronal differentiation and development and it involves also nitration of tyrosine residues in proteins. Although the accumulation of nitrated proteins correlates well with many diseases states, substantial evidence has accrued that protein tyrosine nitration is a post-translational modification playing a role in physiological processes. However, the presence and above all the role of tyrosine nitration in physiological conditions are still controversial. In the present project, we first studied the appearance and localization of nitrotyrosine-containing proteins during neuronal differentiation in PC12 cells and we identified some nitrated cytoskeletal proteins by mass spectrometry.
